Natural cytotoxicity triggering receptor 1 (UniProt: O76036; also known as Lymphocyte antigen 94 homolog, NK cell-activating receptor, Natural killer cell p46-related protein, NK-p46, NKp46, hNKp46, CD335) is encoded by the NCR1 (also known as LY94) gene (Gene ID: 9437) in human. NK-p46 is a single-pass type I membrane protein that belongs to the natural cytotoxicity receptor (NCR) family and serves as a cytotoxicity-activating receptor that may contribute to the increased efficiency of activated natural killer (NK) cells to mediate tumor cell lysis and elimination of virus-infected cells. It is considered as a primary activating receptor mediating NK cell antiviral function. It is involved in the functional recognition of viral HA and hemagglutinin-neuraminidases. NKp46 is shown to be predominantly expressed in the nasal lavage of patients infected with respiratory viruses, but not in normal healthy subjects. Following the initiation of an NK-target cell interaction, NKp46 is reported to cluster at the cell membrane, specifically at the immune synapse where it mediates cytoskeletal rearrangement and cellular polarization. Human NK-p46 is synthesized with a signal peptide of 21 amino acids, which is subsequently cleaved off. The mature form has an extracellular domain (aa 22-258), a helical domain (aa 259-279), and a cytoplasmic domain (aa 280-304). It also has two Ig-like domains (aa 34-118 and 129-211) termed as D1 and D2. Five different isoforms of NKp46 have been described that are produced by alternative splicing. Specificity

Clone 461-G1 specifically recognizes whole human NKp46 and NKp46-D1, but does not recognize NKp46-D2. It targets an epitope with in the first 100 amino acids from the extracellular domain.

Immunogen

Human IgG1 Fc-fused to a recombinant fragment corresponding to the first 100 amino acids from the extracellular domain of human NKp46.

Application

Anti-NKp46 (CD335), clone 461-G1, Cat. No. MABF2059, is a mouse monoclonal antibody that detects Natural cytotoxicity triggering receptor 1 and has been tested for use in ELISA and Flow Cytometry.
