The CLDN4 (claudin 4) gene is mapped to human chromosome 7q11.23. The encoded protein is an integral member, localized to tight junction. It is also expressed in the epithelial basolateral membrane.

Biochem/physiol Actions
CLDN4 (claudin 4) is associated with epithelial differentiation, and is widely implicated in carcinomas. Its expression is considered to differentiate carcinoma with that of sarcoma. Claudin 4 expression is implicated in gastric cancer induced by Helicobacter pylori infection. It contributes to invasion, metastasis and prognosis of the disease. The CLDN4 gene is known to be overexpressed in a number of chemoresistant and metastatic cancer. Its localization to the tight junction prevents paracellular diffusion. Along with this, the extracellular loop interaction mediated by claudin 4 with the neighbouring cells makes it a differentiation marker.
