General description

Kruppel like factor 5 (KLF5) is encoded by the gene mapped to human chromosome 13q22.1. The encoded protein belongs to kruppel-like factor (KLF) protein family and is mainly expressed in variety of tissues, including lung, colon, intestine and pancreas.
Monoclonal Anti-KLF5 (mouse IgG1 isotype) is derived from the hybridoma KLF-55 produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice immunized with a synthetic peptide. KLF5 is a DNA-binding transcriptional regulator. This protein contains three independent peptide modules of the C2H2 zinc finger-type. KLF5 protein is expressed in the prostate, kidney, skeletal muscle, and breast. KLF5 gene is also expressed in embryos and is highly expressed in the later stage of fetal development.

Specificity

Monoclonal Anti-KLF5 recognizes human, monkey, dog, hamster, mouse and rat KLF5.

Immunogen

synthetic peptide corresponding to a sequence at the C-terminus of human KLF5, conjugated to KLH. The corresponding sequence is identical to monkey, bovine, chicken, rat and mouse KLF5.

Application

Anti-KLF5 antibody, Mouse monoclonal has been used in western blotting and immunofluorescence.

Biochem/physiol Actions

KLF5 protein functions through the restoration of cyclin D1, cyclin B1 and Cdc2 gene expression. This protein is also involved in regulating the inhibitory effect of retinoids on cell proliferation. KLF5 protein regulates the pro-proliferative and transforming activities of oncogenic H-ras. KLF5 protein plays a role as a tumor suppressor in intestinal tumorigenesis in vivo. Deletion and down-regulation of KLF5 leads to prostate and breast cancer.
Kruppel like factor 5 (KLF5) has been involved in the regulation of various cellular functions, such as cell cycle, apoptosis, proliferation and differentiation, in both normal and tumor cells. Additionally, it also plays a vital role in maintenance of cardiovascular remodeling. Overexpression of the gene has been observed in various cancer types, including cervical cancer(CC) and colon cancer. Hence, this can be used as a prognostic marker for these cancers.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2-8°C for up to one month. For extended storage, freeze at -20oC in working aliquots. Repeated freezing and thawing,or storage in “frost-free” freezers,is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.
