Considering the significant quenching of fluorescence by manganese (Mn), it is not suitable for the assay. Instead, the use of MgCl2 (up to 1 mM) is suggested as it offers less interference. Additionally, DTT can be used up to 1 mM.

Any challenges encountered with decreases in fluorescence due to buffer components in the assays, such as manganese (1 mM), DTT (5 mM), and the combination of manganese with a reducing agent (both BME and DTT)? It seems that the signal is significantly reduced, if not entirely abolished, when these components are included in the buffer, even with substantial amounts of the PPi standard.
