General description

Protein bicaudal C homolog 1 (UniProt Q9H694; also known as Bic-C) is encoded by the BICC1 (also known as CYSRD) gene (Gene ID 80114) in human. Drosophila bicaudal C and its mammalian homologs are RNA-binding proteins involved in symmetric distribution and localized translation of mRNAs of many proteins in a wide range of cells and tissues. RNA binding of Bic-C is mediated by N-terminal KH domains (a.a. 132-199 & 284-348), whereas a C-terminal sterile alpha motif (SAM; a.a. 873-936) self-polymerizes and localizes Bic-C at cytoplasmic foci where silencing of bound mRNAs takes place. Defective polymerization results in decreased Bic-C stability and thereby attenuates inhibition of Dishevelled-2 of Wnt/β-catenin pathway. Loss of BICC1 provokes renal and pancreatic cysts as well as ectopic Wnt/β-catenin signaling during visceral left-right patterning due to defective silencing of Bic-C target mRNAs, including adenylate cyclase 6 (AC6).

Specificity

Clone 2D9.1 reacts with both spliced isoforms of human and murine BICC1 reported by UniProt (Q9H694 & Q99MQ1).

Immunoprecipitation Analysis: 9 µg from a representative lot immunoprecipitated exogenously expressed human BICC1 isoforms 1 and 2 from 0.5 mL (~50-100 mg) of HEK293 transfectants lysates (Courtesy of Dr. Thomas Tuschl laboratory, The Rockefeller University, New York, NY).

Quality

Evaluated by Western Blotting of recombinant human BICC1.

Western Blotting Analysis: 0.2 µg/mL of this antibody detected 10 µg of recombinant human BICC1.

~130 kDa observed. 104.8/105.0 kDa (Human/mouse isoform 1) and 94.29/93.41 kDa (Human/mouse isoform 2) calculated.
