The recommended blocking agent for PVDF membranes in a Western blot, including when using Casein, as per ther Immobilon®P Transfer Membrane user guide is using a blocking buffer called Block-CH buffer (Cat. No. WBAVDCH01) or a blocking agent such as bovine serum albumin, casein, or nonfat dry milk in wash buffer at a concentration of 0.5-5% (w/v).
Regarding the suitability of Casein as a blocking agent for PVDF membranes, in-house testing has been conducted with IPVH00010 PVDF membrane, and Casein has been found to be suitable. However, it is important to note that users often choose the blocking agent that works best for their specific needs. Therefore, if one prefers to use Casein as a blocking agent with PVDF membrane (IPVH00010), it can be employed as long as it does not interfere with the samples and antibodies being used.
