General description

Lupus Ku autoantigen protein p70 (Ku70) is also known as X-ray repair cross-complementing protein 6 (XRCC6), 5′-dRP lyase Ku70, CTC box-binding factor 75 kDa subunit (CTC75, CTCBF) or Thyroid-lupus autoantigen (TLAA). Ku70 is a single stranded DNA-dependent ATP-dependent helicase that plays a role in chromosome translocation and DNA non-homologous end joining (NHEJ) required for double-strand break repair and recombination. The XRCC5/6 (Ku80/Ku70) dimer acts as regulatory subunit of the DNA-dependent protein kinase complex DNA-PK, and that dimer, together with APEX1, acts as a negative regulator of transcription. Individuals with systemic lupus erythematosus (SLE) and related disorders produce extremely large amounts of autoantibodies to XRCC5 (Ku80) and XRCC6 (Ku70).

Src-family tyrosine kinases are widely expressed in many cell types and participate in a variety of signal transduction pathways. Despite the significance of Src in suppression of apoptosis, its mechanism remains poorly understood. Here we show that Src acts as
