Specificity

Clone 1J15 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Marginal zone B- and B1-cell-specific protein (MZB1).

Immunogen

His-tagged, recombinant full-length human Marginal zone B- and B1-cell-specific protein (MZB1) (without the signal peptide).

Application

Quality Control Testing

Evaluated by Western Blotting in Daudi cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected MZB1 in Daudi cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected MZB1 in Raji, Ramos, PBMC, HEK293T, and HeLa cell lysates.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected MZB1 in human tonsil tissue sections.

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected MZB1 in Raji cells.

Affinity Binding Assay: A representative lot of this antibody bound recombinant human MZB1 protein with a KD of 1.7 x 10-7 in an affinity binding assay.

Target description

Marginal zone B- and B1-cell-specific protein (UniProt: Q8WU39; also known as Mesenteric estrogen-dependent adipose 7, MEDA-7, Plasma cell-induced resident endoplasmic reticulum protein, Plasma cell-induced resident ER protein, pERp1, Proapoptotic caspase adapter protein) is encoded by the MZB1 (also known as MEDA7, PACAP) gene (Gene ID: 51237) in human. MZB1 is a B-cell-specific and endoplasmic reticulum (ER)-localized protein that is implicated in antibody secretion and integrin-mediated cell adhesion. It plays an important role in the humoral immune response by enhancing the interaction of the immunoglobulin (Ig) heavy chain with the chaperone GRP94 and by augmenting the secretion of IgM. It is a part of the ER chaperone complex that contains a large number of molecular chaperones, which associates with unassembled incompletely folded immunoglobulin heavy chains. It associates with immunoglobulin M (IgM) heavy and light chains and promotes IgM assembly and secretion. It is abundantly expressed in innate-like B cells, including Marginal zone (Mz) B cells of the spleen and peritoneal B1 B cells, as well as in antibody-secreting cells. Deletion of MZB1 results in a reduced humoral response with plasma cells secreting lower levels of IgM. Deletion of MZB1 is shown to impair humoral immune responses and antibody secretion in plasma cells. Five isoforms of MZB1 have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
