General description

Ras-related protein Rab-25 (Rab25) is a member of the Rab family of small guanosine triphosphatases (GTPases). The Rab family belongs to the Ras superfamily of small GTPases. Rab25 is specifically expressed in epithelial cells and is localized on human chromosome 1q22. It comprises a GTPase tertiary structure with a central barrel with a six-stranded β-sheet surrounded by α-helices.

Specificity

Anti-Rab25 recognizes human Rab25 (not tested in other species).

Application

Anti-RAB25 antibody produced in rabbit may be used in immunoblotting.

Biochem/physiol Actions

Ras-related protein Rab-25 (Rab25) acts as an oncogene or a tumor suppressor gene. Overexpression of this gene is seen in several cancers such as ovarian cancer, bladder cancer, breast cancer, gastric cancer, liver cancer, non-small cell lung cancer, renal cancer, glioblastoma multiforme and prostate cancer. It helps to control the vesicle trafficking and signaling pathways. Rab25 is associated with the apical recycling system of epithelial cells. Antibodies to Rab proteins may serve as useful tools for studying subcellular localization and membrane organization.

Physical form

Solution in 0.01 M phos­phate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

Store at –20 °C. For continuous use, the product maybe stored at 2–8 °C for up to one month. For extended storage, freeze in working aliquots at –20 °C. Repeated freezing and thawing, or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
