General description

DNAJ/HSP40 family of proteins contains a highly conserved, 70 amino acid J domain that is involved in the interaction with HSP70 and regulation of its ATPase activity. It also contains a glycine and phenylalanine (G/F)-rich region that may function as a flexible linker and a cysteine-rich region (C domain) containing 4 [CXXCXGXG] motifs similar to a zinc-finger domain.

Application

Biochem/physiol Actions

DNAJB12 (DnaJ homolog subfamily B member 12) gene encodes a member of the DNAJ/HSP40 family of proteins that are involved in the regulation of molecular chaperone activity of HSP70 proteins by stimulating ATPase activity. DNAJB12 is a transmembrane protein in the endoplasmic reticulum that functions as a co-chaperone for Hsc70/Hsp70 heat shock proteins, which are involved in the ER-associated degradation of membrane proteins. Overexpression of this protein results in the formation of DJANGOS (DNAJ-associated nuclear globular structures), membranous structures within cell nuclei.
