General description

ColorWheel® technology is a novel and proprietary method of creating your own antibody and dye combinations for use in flow cytometry. By incubating any ColorWheel® antibody with any ColorWheel® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Each ColorWheel® antibody and dye is lyophilized for long-term storage, allowing for simplicity and flexibility without compromise. ColorWheel® technology requires both ColorWheel® antibody and ColorWheel® dye for Flow Cytometry application.

Specificity

Clone 93 is a rat monoclonal antibody that detects CD16/CD32. It targets a conformational epitope formed by FcgII and FcgIII receptors.

Immunogen

VH81X Tg B cells and T220 stromal cells.

Application

Quality Control Testing

Evaluated by Flow Cytometry in BALB/c mouse splenocytes.

Flow Cytometry Analysis: Staining of one million BALB/c mouse splenocytes was performed using 5 μL of a 1:1 mixture of Cat. No. CWA-1085, Anti-Mouse CD16/CD32 (93) ColorWheel® Dye-Ready mAb and Cat. No. CWDS-PE ColorWheel® Antibody-Ready Phycoerythrin (PE) Dye or an equivalent amount of PE-conjugated Rat IgG2a isotype control.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

Target description

Low affinity immunoglobulin gamma Fc region receptor III/II (UniProt: P08508/P08101; also known as IgG Fc receptor III/II, FcRIII/II, CD16/CD32) are encoded by the Fcgr3/Fcgr2 genes (Gene ID: 14131/14130) in murine species. CD16 and CD32 are single-pass type I membrane glycoproteins that are synthesized with a signal peptide (aa 1-30 and 1-29), which is subsequently cleaved off to produce the mature form that contains an extracellular domain (aa 31-215 and 30-210), a transmembrane domain (aa 216-235 and 211-231), and a cytoplasmic domain (aa 236-261 and 232-330). CD16/CD32 are expressed on B cells, monocytes/macrophages, NK cells, granulocytes, mast cells, and dendritic cells. Both CD16 and CD32 can be expressed on the same cell and in varying ratios. They are constitutively expressed, although cytokines and lymphokines can modulate their expression. They serve as low-affinity receptors and bind to the Fc portion of antibody-antigen complexes and play a role in adaptive immune responses. They are involved in a variety of effector and regulatory functions, including phagocytosis of antigen-antibody complexes from the circulation and modulation of antibody production by B-cells. Clone 93 recognizes the common epitope on CD16/CD32.

Physical form

Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.

Reconstitution

1.0 mg/mL after reconstitution at 25 μL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 μL PBS. Protect from light.
