General description

Kallikrein-6 (UniProt Q92876; also known as hK6, Neurosin, Protease M, Serine protease 18, Serine protease 9, SP59, Zyme) is encoded by the KLK6 (also known as PRSS18, PRSS9) gene (Gene ID 5653) in human. Kallikrein-6 belongs to the kallikrein subfamily of serine proteases, members of which are involved in diverse physiological functions and many are used clinically as biomarkers for cancers. Originally identified due to its aberrant expression in mammary and ovarian cancers, kallikrein-6 is expressed in a wide range of tissues, including breast, kidney, and the central nervous system. Kallikrein-6 is known to cleave components of the extracellular matrix (ECM) and is reported to be involved in multiple cellular processes, including inflammation, receptor activation, and apoptosis. In addition, deregulated KLK6 expression occurs in neurodegenerative and skin disorders, and is a common event in many types of human cancers. Consequently, KLK6 represents a promising biomarker of early diagnosis and/or unfavorable prognosis in these human malignancies. Kallikrein-6 is initially produced as a preproprotein with a signal (aa 1-16) and a propeptide (aa 17-21), cleavage of which releases the mature protein (aa 22-244).

Specificity

Clone E24 was raised against the canonical form of human kallikrein-6 recombinant protein. Reactivity against kallikrein-6 spliced isoforms 2 and 3 reported by UniProt (Q92876) has not been determined.

Application

Anti-KLK6 Antibody, clone 27-4 is an antibody against KLK6 for use in Western Blotting, ELISA, Immunoaffinity Purification.
Western Blotting Analysis: 0.5 µg/mL from a representative lot detected kallikrein-6 in 10 µg of human brain tissue lysate.
ELISA Analysis: Representative lots were employed as the capture antibody in combination with biotin-conjugated clone E24 (Cat. No. MABN873) as the detection antibody for analyzing kallikrein-6 levels in ovarian cytosolic extracts as well as ascites fluids from ovarian cancer patients (Shan, S.J., et al. (2007). Br. J. Cancer. 96(2):362-372; Luo, L.Y., et al. (2006). Tumor Biol. 27:227-234).
Immunoaffinity Purification: A representative lot was conjugated to resins and employed for the affinity purification of kallikrein-6 from human cerebrospinal fluids (CSFs) and ascites fluids (Kuzmanov, U., et al. (2009). Mol. Cell. Proteomics. 8(4):791-798).

Quality

Evaluated by Western Blotting in human ovary tissue lysate.

Western Blotting Analysis: 0.5 µg/mL of this antibody detected kallikrein-6 in 10 µg of human ovary tissue lysate.
