General description

Peripheral plasma membrane protein CASK (EC 2.7.11.1; UniProt O14936; also known as Calcium/calmodulin-dependent serine protein kinase, Membrane-associated guanylate kinase, hCASK, Protein lin-2 homolog, Trinucleotide repeat containing 8) is encoded by the CASK (also known as CAGH39, CAMGUK, CMG, FGS4, LIN2, MICPCH, MRXSNA, TNRC8) gene (Gene ID 8573) in human. CASK is a membrane-associated guanylate kinase (MAGUK) family member involved in the regulation of cytoskeletal remodeling and cell migration. It consists of a Ca2+-calmodulin kinase, a PDZ, an SH3, and a guanylate kinase domain. CASK localizes at cell–cell adhesion sites, including synaptic junctions, epithelial junctions, and tight junctions, where it is reported to associate with other cell adhesion molecules, such as junctional adhesion molecule (JAM), cadherins, catenins, and syndecan-2. CASK and syndecan-2 expressions are upregulated in colorectal cancer and their upregulation significantly correlates with advanced tumor stage, lymphatic invasion, lymph node metastasis, vascular invasion, liver metastasis, and unresectable metastatic CRC.
