General description

Nesprin-2 (UniProt: Q6ZWQ0; also known as KASH domain-containing protein 2, KASH2, Nuclear envelope spectrin repeat protein 2, Nucleus and actin connecting element protein, Protein NUANCE, Synaptic nuclear envelope protein 2, Syne-2) is encoded by the Syne2 gene (Gene ID: 319565) in murine species. Nesprin-2 is a single-pass type IV membrane protein with a large cytoplasmic domain (aa 1-6823), a transmembrane domain (aa 6824-2844), and a perinuclear space domain (aa 6845-6874). It contains two calponin-homology domains (aa 31-136 and 183-288) and fifty-six spectrin repeats. Its actin-binding region is localized in amino acids 1-286. Nesprin-2 is a multi-isomeric modular protein that forms a linking network between organelles and the actin cytoskeleton to maintain the subcellular spatial organization. It is a core component of the LINC (LInker of Nucleoskeleton and Cytoskeleton) complex that is involved in the connection between the nuclear lamina and the cytoskeleton. The nucleocytoplasmic interactions established by the LINC complex play an important role in the transmission of mechanical forces across the nuclear envelope and in nuclear movement and positioning. Three different isoforms of Nesprin-2 have been described that are produced by alternative splicing. Different isoform patterns are found in the different compartments of the cell. Nesprin-2 levels are shown to be down-regulated in cancer cells. Nesprin-2 knockdown cells exhibit a defective nuclear translocation of c-Fos and Smad 2/3. Mutations in Syne2 gene have been linked to Emery-Dreifuss muscular dystrophy 5 (EDMD5), an autosomal dominant form of degenerative myopathy. (Ref.: Zhang, Q., et a; (2019). Curr. Specificity

This rabbit polyclonal antibody detects Nesprin-2. It targets an epitope within the N-terminal, actin-binding region.

Immunogen

GST-tagged recombinant fragment corresponding to 284 amino acids from the N-terminal, actin-binding region of mouse Nesprin-2.

Application

Quality Control Applications

Evaluated by Immunocytochemistry in NIH3T3 cells.

Immunocytochemistry Analysis: A 1:1,000 dilution of this antibody detected Nesprin-2/SYNE2 in NIH3T3 cells.

Tested applications

Western Blotting Analysis: A representative lot detected Nesprin-2/SYNE2 in Western Blotting application (Luxton, G.W., et. al. (2010). Science. 329(5994); 956-9).

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Nesprin-2/SYNE2 in NIH-3T3 overexpressing Nesprin-2.

Immunocytochemistry Analysis: A representative lot detected Nesprin-2/SYNE2 in immunocytochemistry application (Zhang, Q., et. al. (2019). Curr Biol. 29(17); 2826-2839).

Storage and Stability

Store at -10°C to -25°C. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
