General description

Amyloid-beta A4 precursor protein-binding family B member 1 (UniProt: Q9QXJ1; also known as Protein FE65) is encoded by the Apbb1 (also known as FE65) gene (Gene ID: 11785) in murine species. FF65 is an adapter protein that forms a transcriptionally active complex with the gamma-secretase-derived amyloid precursor protein (APP) intracellular domain. It is selectively expressed in the brain with high expression in regions affected in Alzheimer s disease. FE65 is phosphorylated at Threonine 579 by GSK-3 beta, which potentiates gamma and beta secretase-mediated APP processing and beta-amyloid liberation. This phosphorylation is also reported enhance FE65/APP complex formation. FE65 also plays a central role in the response to DNA damage by translocating to the nucleus and inducing apoptosis. FE65 may act by specifically recognizing and binding histone H2AX phosphorylated on Tyrosine 142 (H2AXY142ph) at double-strand breaks, recruiting other pro-apoptosis factors such as MAPK8/JNK1. FE65 contains an N-terminal WW domain (aa 253-285) and two C-terminal phosphotyrosine interaction domains (aa 370-509, PID1 and aa 542-699, PID2). (Ref.: Lee, YS et al. (2017). Sci. Specificity

Clone 3H6 specifically detects Protein FE65 in murine and human cells.

Immunogen

Epitope: cytoplasmic domain
GST-tgged recombinant fragment corresponding to 40 amino acids from the N-terminal half of murine Protein FE65.

Application

Anti-FE65, clone 3H6, Cat. No. 05-758-I, is a highly specific mouse monoclonal antibody that targets FE65 and has been tested for use in Western Blotting.
