Neutrophil elastase (UniProt: P08246; also known as EC:3.4.21.37, Bone marrow serine protease, Elastase-2, Human leukocyte elastase, HLE, Medullasin, PMN elastase) is encoded by the ELANE (also known as ELA2) gene (Gene ID: 1991) in human. Neutrophils are the predominant circulating leukocyte population and are important in modulating innate and adaptive immunity. They serve as early responders and are recruited to sites of inflammation and infection by environmental cues. Neutrophil elastase (NE), a member of the peptidase S1 family, is synthesized with a signal peptide (aa 1-27) and a short propeptide (aa 28-29) that are subsequently cleaved off to produce the mature NE (aa 30-267). The mature form contains two glycans and four disulfide bonds. Release of NE can also stimulate adverse remodeling by degrading virtually all the components of the ECM in addition to elastin, and including collagen, fibronectin, and laminin. It is reported to cleave the triple-helix structure of type III and IV collagens, and cross-linked regions of type I, II, and V collagens, aggrecans, and fibronectin. NE-induced degradation of extracellular matrix (ECM) can release bioactive peptides and growth factors such as EGF and FGF that have mitogenic and motogenic effects on smooth muscle cells and fibroblasts. Increased activation of NE can lead to activation of matrix metalloproteinases that could potentiate ECM degradation. NE is also reported to cleave CXCL12, a chemokine involved in the regulated release of neutrophils from the bone marrow. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
