Signal transducer and activator of transcription 1-alpha/beta (UniProt: P42224; also known as Transcription factor ISGF-3 components p91/p84, STAT1) is encoded by the STAT1 gene (Gene ID: 6772) in human. STATs (signal transducers and activators of transcription) are a family of cytoplasmic latent transcription factors that are activated to regulate gene expression in response to a large number of extracellular signaling proteins. STAT1 is activated by many different ligands including interferons, interleukins, and growth factors. STAT1 exists in two forms, the 91 kDa STAT1alpha and the 84 kDa STAT1beta, which are encoded by the same gene with splicing variant. Following ligand binding to cell surface receptors, signaling via protein kinases leads to activation of Jak kinases and tyrosine phosphorylation of STAT1 and STAT2. The phosphorylated STATs dimerize and enter the nucleus. Phosphorylation on serine 727 by several kinases including MAPK14, ERK1/2 and CAMKII following on IFN-gamma stimulation, regulates STAT1 transcriptional activity. This phosphorylation also promotes sumoylation though increasing interaction with PIAS. Application

Rabbit recombinant monoclonal antibody Anti-STAT1 (isoform ), clone 1D4 ZooMAb Rabbit Monoclonal, Cat. No. ZRB1459 is used in Western Blotting, Immunohistochemistry (Paraffin), Immunocytochemistry and Affinity Binding Assay, for the detection of STAT1.

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected STAT1 in HEK293 cell lysate.

Immunocytochemistry Analysis: A 1:100-1,000 dilution from a representative lot detected STAT1 in U2OS and Hela cells treated with interferon-gamma.

Immunohistochemistry (Paraffin) Analysis: A 1:100-1,000 dilution from a representative lot detected STAT1 in human kidney and human breast cancer tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound recombinant STAT1 alpha fragment with a KD of 1.1 x 10-6 in an affinity binding assay.

Biochem/physiol Actions

Clone 1D4 is a a ZooMAb rabbit recombinant monoclonal antibody that specifically detects human STAT1. It targets an epitope within 38 amino acids from the C-terminal region.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in from PBS with 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Preparation Note

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.

Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
