General description

Clusterin (UniProt: P10909; also known as Aging-associated gene 4 protein, Apolipoprotein J, Apo-J, Complement cytolysis inhibitor, CLI, Complement-associated protein SP-40,40, Ku70-binding protein 1, NA1/NA2, Sulfated glycoprotein 2, SGP-2, Testosterone-repressed prostate message 2, TRPM-2) is encoded by the CLU (also known as APOJ, CLI, KUB1, AAG4) gene (Gene ID: 1191) in human. Clusterin is a ubiquitously expressed protein that functions as extracellular chaperone that prevents aggregation of non-native proteins. It also prevents stress-induced aggregation of plasma proteins. Clusterin is synthesized with a signal peptide (aa 1-22), which is subsequently cleaved off to generate the mature form that can be proteolytically cleaved into a b-chain (aa 23-227) and an a-chain (aa 228-449). However, proteolytic cleavage is not necessary for its chaperone activity. Six different isoforms of Clusterin have been described that are produced by alternative splicing. Isoform 1 is shown to retro-translocate from the secretory compartments to the cytosol upon cellular stress, but isoforms 4 and 6 retain their cytoplasmic localization in stressed and unstressed cell. It maintains partially unfolded proteins in a state appropriate for subsequent refolding by other chaperones, such as HSPA8/HSC70, but does not refold proteins by itself. Clusterin is reported to bind to Amyloid b-peptides and alters their aggregation and promote their clearance, suggesting its neuroprotective role. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Immunofluorescence Analysis: A 1:100 dilution from a representative lot detected Clusterin a in human tonsil tissue sections.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Clusterin a in human adrenal gland and human spleen tissue sections.

Biochem/physiol Actions

Clone 41D is a ZooMAb® mouse recombinant monoclonal antibody that specifically detects human Clusterin a.

Physical form

Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb® formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Preparation Note

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
