General description

Factor Xa (FXa) is the activated form of the coagulation factor X (Stuart-Power factor, thrombokinase, prothrombinase, thromboplastin, E.C.3.4.21.6). Factor X is a vitamin K-dependent serine endopeptidase plays an important role at several stages of the coagulation pathway. It is produced in the liver and in the plasma, it circulates as a two-chain glycoprotein. It acts by converting prothrombin into active thrombin by complexing with activated co-factor V in the prothrombinase complex. Unfractionated heparin and various low molecular weight heparins bind to plasma cofactor antithrombin to inactivate several coagulation factors including factor Xa.

Application

Suitable for the determination of Factor Xa activity in plasma and purified Factor Xa samples.

Biochem/physiol Actions

This Factor Xa activity assay kit utilizes the ability of Factor Xa to cleave a synthetic substrate thereby releasing a fluorophore, AMC, which can be quantified by fluorescence readers. This assay kit is simple, rapid, and can detect activity from as low as 1 ng of Factor Xa.

Questions

1–2 of 2 Questions
  1. Does it matter what kind of plasma? EDTA, citrate?

    1 answer
    1. For plasma samples, citrated plasma or heparinized plasma is recommended, as EDTA may interfere with the enzymatic assay.

      Helpful?

  2. Is the factor Xa activity kit (MAK238) suitable for mouse plasma samples? Is it specific for human, or can it also be used with mouse samples?

    1 answer
    1. The kit can be used for plasma samples, and it has been tested with a synthetic material rather than with human or mouse plasma. The assay uses a synthetic substrate to assess Factor Xa activity, indicating that the kit should technically be suitable for use with any plasma samples. The reaction involves the cleavage of FXa Substrate-AMC by FXa enzyme, resulting in cleaved Substrate + AMC (Fluorescence). Its applications include determining the activity of pure Factor Xa and detecting the activity of Factor Xa in plasma.
