Application

Lipopolysaccharides from Escherichia coli O111:B4 has been used:
  • to stimulate an inflammatory response in porcine liver tissues in-vivo and piglet ileum tissues ex vivo
  • to stimulate Ex vivo natural killer(NK) cells and hepatic B cells[1]
  • to induce arthritis in a standard rabbit model

Lipopolysaccharides (LPSs) are characteristic components of the cell wall of Gram-negative bacteria. LPS and its lipid A moiety stimulate cells of the innate immune system by the Toll-like receptor 4 (TLR4), a member of the Toll-like receptor protein family, which recognizes common pathogen-associated molecular-patterns (PAMPs).

Biochem/physiol Actions

Lipopolysaccharides (LPS) are distributed on the outer layer of the membrane and in non-capsulated strains, present on the cell surface. LPS maintains the outer membrane integrity. In addition, it protects the cell from bile salts stress and lipophilic antibiotics.

Preparation Note

The product is soluble in water (5 mg/ml) or cell culture medium (1 mg/ml) yielding a hazy, faint yellow solution. A more concentrated, though still hazy, solution (20 mg/ml) has been achieved in aqueous saline after vortexing and warming to 70-80 oC. Lipopolysaccharides are molecules that form micelles in every solvent. Hazy solutions are observed in water and phosphate buffered saline. Organic solvents do not give clearer solutions. Methanol yields a turbid suspension with floaters, while water yields a homogeneously hazy solution.

  1. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  2. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers.   3. What is recommended for the storage of longer duration once opened? Is it better to keep at 2°C-8°C for 1-2 years or to freeze it (-80°C or -20°C) and thaw it gently when needed?

    1. This product is stable at 2-8°C at least until the assigned retest date, provided that no contaminants have been introduced. The recommended retest date is lot specific and reported in the product Certificate of Analysis. The stability of the product in dilute form or beyond the assigned retest date has not been determined. However, LPS products prepared from the powder form may be stored in 1 mg/mL aliquots for up to 2 years at -20°C. The storage of solutions at -80°C has not been determined.
