General description

10% Neutral buffered formalin (NBF) is the most commonly used fixative throughout the world for light microscopy and is a somewhat forgiving fixative. 10% NBF is well suited for large throughput laboratories, and requires a relatively short period of fixation, but can also be used for the long-term storage of tissue. Formalin fixation is thought to form between the aldehydes and the proteins, creating a gel, thus retaining cellular constituents in their in vivo relationship. Once properly fixed, the tissue should be able to withstand the subsequent stages of tissue processing or staining.

Application

Formalin solution, neutral buffered, 10% has been used in oil-red-o staining for the differentiation of 3T3-L1 preadipocyte cell line and mouse embryonic fibroblasts into adipocytes. It has been used in the histological analysis of rabbit′s eye and diabetic rat intestine.[1]

    1. The exact buffer concentration for HT511128 is not stated on the website and is considered confidential. The classic textbook formulation for 10% Neutral Buffered Formalin is as follows:

      100 ml Formaldehyde, 37-40%
      900 ml distilled water
      4.0 grams sodium phosphate, monobasic
      6.5 grams sodium phosphate, dibasic (anhydrous)

      The Specification Sheet states the pH range must be between 6.90 to 7.10. The temperature at which the pH is not measured but should be either 20° or 25°C.

Osmolality is significant for live cells, as cells struggle to survive when their surroundings are vastly different from their own, on the other hand formalin is only a fixative solution. Formaldehyde solutions are buffered to prevent the formation of formalin pigment or acid hematin that forms when tissues are fixed at low pH.

With HT501128, the formaldehyde used for dilution has a formaldehyde content of 37 to 40%. When the product is diluted 1 to 10 with water, it is referred to as 10% formalin, which has a formaldehyde content of 4% formaldehyde. Both products have the same concentration of formaldehyde in solution.
