General description

Bovine serum albumin is a model protein in various studies and is made up of 583 amino acids. It is a water-soluble protein with a molecular weight of 66.4kDa. Six α-helices form three homologous domains of BSA. Depending on pH, it undergoes reversible conformational isomerization. The native structure of the protein becomes reactive and flexible on heating.

Application

Bovine Serum Albumin has been used in the washing solution for flow cytometry analysis, tissue digestion solution and blocking solution for ELISA (enzyme-linked immunosorbent assay). It has also been used in the resuspension buffer for flow cytometry.

Biochem/physiol Actions

Bovine serum albumin is broadly used as an additive to cell culture media, especially serum-free media. It provides a range of benefits including protection from oxidative damage and stabilization of other media components such as fatty acids and pyridoxal.
Certain conformational and primary-sequence epitopes of BSA are suspected allergens in human beef and milk allergies.

Preparation Note

Often referred to as Cohn fraction V; this product is prepared by a modified method of the Cohn cold ethanol fractionation method.
Serum albumin may be referred to as Fraction V. This naming convention is taken from the original Cohn method of fractionating serum proteins using cold ethanol precipitation. Serum albumin was found in the fifth ethanol fraction using Cohn′s method. Since then, the term "Fraction V" has been used by some to describe serum albumin regardless of the method of preparation. Others have used this term to describe serum albumin purified by ethanol fractionation methods that have been highly modified since the original Cohn method was described. Sigma-Aldrich manufactures and distributes serum albumins purified from a variety of primary methods including the true Cohn fractionation method, modified ethanol fractionation methods, heat shock and chromatography. Additional purification steps may include crystallization or charcoal filtration.

  1. Is BSAS A9418, Protease-free and suitable for Organoid Culture (Growth Factor preparations)? This Cat# doesn't say Fraction-V? Is it protease free or not?

    1. This product is Cohn fraction V Albumin prepared by a modified method of Cohn cold alcohol precipitation. It is not tested for protease content. It is suitable for general cell culture applications, so it is expected to be suitable for organoid culture.   2. Hello, Can you send me the amino acid sequence and the Uniprot or PDB file?

    1. Historically, the molecular weight of BSA has been cited as 66,1201 or 66,2672. However, it was revised in 1990 to 66,4303. All three values are based on amino acid sequence information available at the time of publication. The complete sequence can be found in the UniProt

  3. I make a mistake to put this BSA powder  (A9647-100G) in the room temperature for 39 hours. Can it still work?

    1. The long term storage recommendation for this product is refrigerated at 2-8 °C, but it is shipped at ambient temperature. Refrigerated products that ship at ambient temperature are stable for up to 2 weeks to allow for transit time. Items left at room temperature for short periods will not be adversely effected.

  4. Would it be safe to use product A9418 if it was accidentally stored at -20°C for approximately 24 hours, even though it is intended to be stored at 2-8°C?

    1. The performance of the item after storing it at -20°C cannot be guaranteed, and testing its functionality is recommended.

  5. Can BSA ever contain alpha1 anti-trypsin as a contaminant?

    1. The available Bovine Serum Albumin products are not tested for the presence of alpha-1 antitrypsin (AAT). AAT is typically found in Bovine Serum, however, it is unknown if the isolation and purification methods employed in the production of BSA would degrade this protein. A brief search of the literature shows AAT has a molecular weight range of 45 - 52 kDa. It is irreversibly degraded at pH levels below 5. A further review of the literature on potential removal or purification would be recommended.
