General description

We are committed to bringing you Greener Alternative Products, which adhere to one or more of The 12 Principles of Greener Chemistry. This product has been enhanced for energy efficiency and waste prevention when used in starch hydrolysis research. For more information see the article in biofiles.

Amyloglucosidase is a disaccharide enzyme belonging to the α-glucosidase type. It is produced by several species of Aspergillus genus.

Application

Amyloglucosidase from Aspergillus niger has been used in glycogen assay and in acetate buffer for starch hydrolysis.[1] It can also be used for glycogen determination in whole yeast cells and synthesis of hetero-oligosaccharides by glucoamylase in reverse.

Other Notes

One unit corresponds to the amount of enzyme which liberates 1 μmole of glucose per minute at pH 4.8 and 60 °C (starch acc. to Zulkowsky, Cat. No. 85642, as substrate).

Questions

1–3 of 3 Questions
  1. What is the optimal temperature for this enzyme?

    1 answer
    1. The enzymatic activity of this product is assessed at 60 °C with glucose as a substrate. External sources indicate higher temperatures, up to 75°C, may be optimal for more complex sugars.

      Helpful?

  2. Can you kindly indicate what the optimum pH is for this enzyme?

    1 answer
    1. The activity of this enzyme is assessed at pH 4.8 with glucose as a substrate. External sources indicate a lower pH of approximately 4.0 may be optimal for a disaccharide, such as maltose, or a more complex sugar.

      Helpful?

  3. What enzyme dilution buffer is recommended for this specific enzyme?

    1 answer
    1. For assay purposes, this product is prepared in water acidified to pH 4.8 at a concentration of 5 u/mL. This material may also be prepared in 10 mM sodium acetate buffer, pH 4.8.

      Helpful?
