Opis

General description

Bismuth Sulfite Agar is a highly selective medium designed for the isolation and detection of Salmonella species, including Salmonella typhi, from food, water, and clinical samples. The medium′s selectivity is achieved through the combined action of freshly precipitated bismuth sulfite and brilliant green, which effectively suppresses the growth of coliforms while permitting the growth of salmonellae. Sulfur compounds in the medium provide a substrate for hydrogen sulfide production, and the metallic salts, stain the colonies and surrounding medium black or brown in the presence of hydrogen sulfide, providing a clear visual indicator for Salmonella identification.

Application

Bismuth Sulfite Agar is used for the selective isolation and detection of Salmonella species in food, water, and clinical specimens. The medium is particularly valuable in food safety and clinical microbiology laboratories where the detection of Salmonella is critical. When the sample contains high levels of organic matter, it is recommended to suspend the sample in sterile saline and use the supernatant for inoculation to prevent the formation of atypical colonies.

Preparation Note

Dissolve 47.55 g in litre distilled water and heat to boiling to dissolve the medium completely. DO NOT STERILIZE. This medium should be prepared, and the plates poured, 24h in advance of their intended use, since the freshly prepared medium is excessively toxic to Salmonellae. It must also be remembered the medium loses selectivity after 3-4 days, even when refrigerated.
