The signal from staining is generally maintained after formaldehyde fixation. However, prolonged storage beyond 24 hours may result in some signal attenuation, depending on storage conditions. For optimal retention, store samples at 2 to 8°C and protect them from light.

General description
The BioTracker 650 Red Nuclear Dye is a cell-membrane permeable red fluorescent DNA dye that specifically stains nuclei in live or fixed cells. It has excellent specificity for DNA without the need for a wash step, and it has low toxicity for live cell imaging. The dye is supplied with a vial of verapamil, an efflux pump inhibitor that may improve probe retention and live cell staining in certain cell types.

Is there a risk of interference from phenol red in cell culture medium with the detection of this dye, as the protocol recommends dilution in cell culture medium?

Phenol red is unlikely to interfere with the detection of this dye. However, it's unnecessary to have phenol red in the cell culture medium at this point. Highly recommend using cell culture medium without phenol red.

Is this dye compatible to image live migrating HL60 cells?

This product is suitable for live cell imaging. The HL60 cell line has not been specifically tested, however there is no reason to expect it will not work. While this stain is markedly more photostable that DAPI and other similar dyes, the retention time has not been determined. The addition of verapamil is recommended to improve retention time.
