Opis

General description

Serine/threonine-protein kinase N2, also known as Protein kinase C-like 2 or Protein-kinase C-related kinase 2, and encoded by the gene name PKN2/PRK2/PRKCL2, is a PKC-related serine/threonine-protein kinase and Rho/Rac effector protein that participates in specific signal transduction responses in the cell. PKN2 has been shown to play an important role in the regulation of cell cycle progression, actin cytoskeleton assembly, cell migration, cell adhesion, tumor cell invasion and transcription activation signaling processes. Protein-kinase-C-related kinases (PRKs) are part of the lipid-regulated protein kinases (PKC) which also include liver PAK & PKN. Human PRK1 and PRK2 share structurally similar catalytic domains, but less similar N-terminal regulatory regions suggesting different regulatory domain functions. PRK1 and PRK2, as well as a third member of this family, PRK3, show distinct patterns of expression in adult tissues. Additionally, the serine-threonine kinase PRK2 can be specifically cleaved by caspase-3 (and/or caspase-3-like subfamily members) during apoptosis. PKN2 is expressed in numerous tumor cell lines, especially in bladder tumor cells.

Anti-PKN2, clone 1D1 Antibody is an antibody against PKN2 for use in western blotting, ELISA, IHC, flow cytometry.

ELISA: A representative lot detected control antigen (100 ng) and antigen (10 ng, 50 ng, 100 ng) in ELISA (Direct).

Immunohistochemistry Analysis: A 1;200-1,000 dilution from a representative lot detected PKN2 in prostate and lung cancer tissues.

Flow Cytometry Analysis: A 1:200-400 dilution from a representative lot detected PKN2 in NIH/3T3 cells.

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.

Analysis Note

Evaluated by Western Blotting in PC-12, Cos7, K562, Jurkat, HeLa, A431, C6, NIH/3T3 and HEK293 cell lysates.

Western Blotting Analysis: A 1:500-2,000 dilution of this antibody detected PKN2 in PC-12, Cos7, K562, Jurkat, HeLa, A431, C6, NIH/3T3 and HEK293 cell lysates.
