Ribonuclease A, Protease-Free is a

chromatographically purified, pyrimidine-specific

endoribonuclease that acts on single-stranded

RNA.
Ribonuclease A, Protease-Free is achromatographically purified, pyrimidine-specificendoribonuclease that acts on single-strandedRNA.

Preparation Note

Following reconstitution, aliquot and freeze (-20°C). Stock solutions are stable for up to 6 months at -20°C.

Analysis Note

May contain trace amounts of DNase I, which can be inactivated by boiling for 15 min in 50 mM potassium acetate, pH 5.5.

Other Notes

One unit is defined as the amount of enzyme that will catalyze the hydrolysis of RNA to yield a first-order velocity constant equal to 1 at 25°C, pH 5.0.

  1. What is the effectiveness of RNAseA when used with a buffer containing 500mM NaCl?

    1. Our laboratory has not specifically studied the impact of high salt, such as 500 mM NaCl, on the activity of RNAseA. However, Park and Raines (2003) have investigated the activity of RNAseA under high salt concentration, up to 1 M NaCl. The results available in the publication titled "Catalysis by ribonuclease A is limited by the rate of substrate association" may serve as a useful reference. The study is available in Biochemistry, 2003 Apr 1;42(12):3509-18. From the study, it is noted that "the pKa values at 1.0 M NaCl are expected to be higher by 0.4-0.5 unit than the pKa values determined at 0.142 M NaCl."
