General description

This panel targets phosphorylated and total Akt1, a key isoform involved in regulating cell proliferation, metabolism, and survival. Akt1 (pS473) is activated downstream of PI3K and is implicated in cancer, cardiovascular disease, and metabolic disorders. The panel enables isoform-specific analysis of Akt signaling.

Application

MILLIPLEX® Qualified assays undergo rigorous assay development, verification, and Quality Control testing to achieve optimal performance. Simultaneously analyze 2 analytes in human, mouse, or rat cell lysate samples.

Questions

  1. How does this assay work? Will the presence of a common antibody between the two beads impact the results? Specifically, will pAKT bind to the total beads and will total AKT (not phospho) bind to the pAKT beads?

    1. In the case of phospho and total 2plex, the capture/bead is specific, while the detection is common. Therefore, the phospho capture/bead should exclusively capture the phosphorylated protein, while the total capture/bead will capture all remaining non-phosphorylated proteins.

      In theory, an increase in the phospho signal would likely cause a slight drop in the total protein signal. However, for every phospho/total 2-plex, a comparison between singleplex and multiplex is always conducted to demonstrate that both phospho and total signals are similar, regardless of whether they are run as singleplex or multiplex.
