Application

Lissamine Green B has been used in blood-brain barrier (BBB) studies and is used to study the its effect on herpes simplex virus 1 (HSV-1) corneal infection in animals.

Biochem/physiol Actions

Lissamine green also referred to as acid green 50, wool green S, food green S or green S, is used for vital staining of cornea and conjunctiva. Staining characteristics are almost similar to rose bengal dye. Lissamine green is also used to stain membrane-damaged epithelial cells. In vitro, Lissamine green B at low concentration blocks herpes simplex virus 1 (HSV-1) plaque formation.

    1. The Sigma Aldrich Handbook of Stains, Dyes and Indicators states Lissamine Green B has been used to dye wool, silk (from a sulfuric acid bath), to print wool and silk directly and to coat paper, dye leather and surface stain wood.

    1. The Sigma Aldrich Handbook of Stains, Dyes and Indicators advises that Lissamine Green B has been substituted for Fast Green FCF without any real advantage. Lillie found it performed well as a collagen stain in Masson's trichrome and as a counter stain in the Van Gieson method. However, it was inferior when used as a component in Van Gieson's Trichrome Stain. See also Clark G, Staining Procedures, 4th Edition. (Williams & Wilkins, Baltimore, 1981), p. 120 for the Lillie modification of Masson's trichrome method.  This material is used in the determination of chlorine dioxide in solution in EPA Method 32.

    1. Lissamine Green B is soluble in water up to 0.1% in water. Other sources have stated solubility for some lots can approach 4%. Solubility in ethanol is 3 mg/mL, although this is not tested by Sigma Aldrich on a lot to lot basis. Lissamine Green B has also been reported to be soluble in Cellosolve, and in ethylene glycol.
