Specificity

Clone 3I3-L2 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects FGF-20. It targets an epitope within 24 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 24 amino acids from the N-terminal half of human FGF-20.

Application

Tested Applications

Affinity Binding Assay: A representative lot of this antibody bound FGF-20 peptide with a KD of 1.0 x 10-12 in an affinity binding assay.

Target description

Fibroblast growth factor 20 (UniProt: Q9NP95; also known as FGF-20) is encoded by the FGF20 gene (Gene ID: 26281) in human. FGF-20 is a homodimeric protein that serves as a neurotrophic factor and regulates the development and function of central nervous system. It is predominantly expressed in the cerebellum and substantia nigra. Human and rat FGF-20 display high degree of homology (~ 95%). It acts on proximal cells and significantly enhances the survival of dopaminergic neurons by activating the MAPK pathway through FGF receptor 1c. In the rat model of Parkinson′s disease, FGF-20 is shown to protect against the loss of dopaminergic neurons. It is shown to ACT® synergistically with FGF-2 to increase the number of dopaminergic neurons in primate embryonic stem cell–derived neurospheres composed of neural progenitors. Exposure to FGF-20, along with Wnt5a and FGF-2, during the final stage of in vitro differentiation, can enhance the maturation and in vivo survival of dopaminergic neurons. Mutations in FGF20 gene have been linked to Renal hypodysplasia/aplasia 2 (RHDA2), a perinatally lethal renal disease with kidney development defects, bilateral renal aplasia, hypoplasia, and severe obstructive uropathy. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, Affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
