General description

ULK1 (unc-51 like autophagy activating kinase 1) is a Ser/Thr protein kinase which forms a component of the ULK1 protein kinase complex. This protein complex also contains the adaptor proteins ATG (autophagy-related) 13, RB1CC1/FIP200 (RB1-inducible coiled-coil 1), and ATG101. ULK1 protein is one of the five orthologs of yeast Atg1 found in mammals.

Biochem/physiol Actions

ULK1 (unc-51 like autophagy activating kinase 1) protein kinase plays a crucial role in the control of autophagy initiation. This protein is responsible for activating a cascade of signaling pathways which ultimately results in the formation of phagosomes. The phosphorylation of ULK1 by MTORC1 (mammalian target of rapamycin complex) leads to the negative regulation of autophagy. Activation of type I interferon (IFN) receptor (IFNR) results in the activation of ULK1 protein, which, in turn, results in the transcription of gene in a IFN-stimulated response elements (ISRE) and IFNγ activation site (GAS) elements-mediated manner. It is also required for the control of the expression of key IFN-stimulated genes (ISGs).

Physical form

Formulated in 2 5mM Tris-HCl, pH 7.5, 100 mM NaCl, 0.05% Tween-20, 100 μg/ml FLAG peptide, 20% glycerol and 3 mM DTT.

Preparation Note

Thaw on ice. Upon first thaw, briefly spin tube containing enzyme to recover full content of the tube. Aliquot enzyme into single use aliquots. Store remaining undiluted enzyme in aliquots at -70°C. Note: Enzyme is very sensitive to freeze/thaw cycles.
