Biochem/physiol Actions

Windorphen is a selective Wnt/β-catenin signaling inhibitor acting through selective inhibition of p300, a histone acetyltransferase (HAT) that also acts as a critical coactivator of β-catenin, a transcription factor that mediates the Wnt signaling pathway involved in embryogenesis and cancer cell survival. Windorphen blocks Wnt signaling by selectively disrupting the association of p300 with the C-terminal transactivation domain of β-catenin-1. Windorphen does not inhibit the closely related CREB-binding protein (CBP). Windorphen induced apoptosis in several Wnt-dependent tumor cell lines including colon adenocarcinoma SW480and RKO and prostate cancer cell lines DU145 and PC3.
