General description

Alcohol oxidase (AOX) is a homo-octamer, which has a molecular weight of 600-kDa. It has eight flavin adenine dinucleotide (FAD) cofactors. It belongs to the glucose-methanol-choline (GMC) oxidoreductase family.

Application

Alcohol Oxidase may be used to study protein translocation into peroxisomes. Product A2404 is from Pichia pastoris. It has been used for the bacterial expression and immunological verification of Hv-p68 cDNA clones .
Alcohol Oxidase solution from Pichia pastoris has been used in cotton pectin methylesterase (PME) enzyme assay. It has also been used to immobilize onto polyaniline (PANI) film as a visual biosensor in dip-stick format for the determination of ethanol in fermented beverage samples.

Biochem/physiol Actions

Alcohol oxidase catalyzes the oxidation of short-chain, primary, aliphatic alcohols to their respective aldehydes. It has the highest affinity for methanol. Alcohol oxidase is involved in the metabolism of methanol, which is important for yeasts that utilize methanol as their sole energy source. Alcohol oxidase is primarily found in the peroxisome but may be present in the cytoplasm as well.

Unit Definition

One unit will oxidize 1.0 μmole of methanol to formaldehyde per min at pH 7.5 at 25 °C.

Protocols

To measure alcohol oxidase activity, this assay uses 2,2′-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) and a continuous spectrophotometric rate determination at 405 nm.
