General description

Transcription intermediary factor 1-alpha (TIF1-alpha) is also called E3 ubiquitin-protein ligase TRIM24, RING finger protein 82, and Tripartite motif-containing protein 24. TIF1-alpha is a transcriptional coactivator that interacts with chromatin depending on histone H3 modifications, with the highest affinity for histone H3 that is both unmodified at ′Lys-4′ (H3K4me0) and acetylated at ′Lys-23′ (H3K23ac). TIF1-alpha has E3 protein-ubiquitin ligase activity, helps regulate cell proliferation and apoptosis and up-regulates transcription activation. Aberrant TIF1-alpha can cause thyroid papillary carcinoma (TPC) and is up-regulated in some breast cancers.

Specificity

Other homologies: Rat (93% sequence homology).
This antibody recognizes the C-terminus of the Nuclear Receptor Binding Site (NRBS) of TIF1-alpha.

Immunogen

Epitope: C-terminus of the Nuclear Receptor Binding Site (NRBS)
KLH-conjugated linear peptide corresponding to the C-terminus of the Nuclear Receptor Binding Site (NRBS) of human TIF1-alpha.

Application

Detect TIF1α using this rabbit polyclonal antibody, Anti-TIF1-alpha Antibody validated for use in western blotting & ICC.
Immunocytochemistry Analysis: A 1:50 dilution from a represenative lot detected TIF1-alpha in HeLa cells.

Quality

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: 0.1 µg/mL of this antibody detected TIF1-alpha in 10 µg of HeLa cell lysate.

Target description

~140 kDa observed. Uniprot descrives 2 isoforms at ~117 kDa and ~113 kDa due to alternative splicing. This protein has been observed at ~140 kDa (Gunawardena, H., et al. (2008). Rheumatology (Oxford). 47(3):324-328.).
