General description

Fibroblast activation protein alpha (FAP), also known as Seprase, is a 97 kDa, type II cell surface glycoprotein belonging to the serine protease family. Although FAP is considered fibroblast specific, it is not found in normal healthy fibroblasts. FAP is most commonly found in stromal, tumor fibroblasts, and is likely involved in ECM pericellular proteolysis, endothelial cell migration and invasion into the ECM. The presence of FAP in human epithelial carcinoma is increased, particularly in breast, colorectal, lung, and ovarian carcinomas. FAP is also transiently observed in developing cells, or in tissues that are undergoing wound healing or suffering from chronic inflammatory conditions. Due to its specific distribution both to tumor fibroblasts and to inflammed tissue, FAP makes a promising target for research in tumor-stromal studies and in therapeutic studies.

Application

Detect Fibroblast activation protein alpha (FAP) using this Anti-Fibroblast activation protein alpha (FAP) Antibody validated for use in WB, IH.
Immunohistochemistry: A 1:750 dilution of a previous lot of FAP antibody was used to immunohistochemically stain frozen tissue sections of HEK-FAP or HT-29 xenograft tumors from C.B17 mice (Cheng, 2002).

Inhibition: Inhibition of dipeptidyl peptidase activity with a previous lot of FAP antibody on HT-29 xenograft tumors was demonstrated by an independent laboratory (Cheng, 2002).

Evaluated by Western Blot in HEK-FAP xenograft tumor lysate.

Western Blot Analysis: 2.5 µg/mL of this antibody detected FAP in 10 µg of HEK-FAP xenograft tumor lysate.

Target description

~ 90 kDa observed MW. Two isoforms of the Fibroblast activation protein exist at calculated MWs of 88kDa and 27 kDa It is possible that both bands may be obserbved in certain cell lines.
