General description

Facade-EM is a two-dimensional amphiphile with hydrophobic and hydrophilic faces that has superior ability to stabilize integral membrane proteins. This new class of detergents represents a departure from traditional one-dimensional polar-head/nonpolar-tail molecules.

Application

FacadeEM (3α-hydroxy-7α,12α-di-((O-β-D-maltosyl)-2-hydroxyethoxy)-cholane) may be used:
  • as a facial amphiphile detergent to purify protoxin-II (ProTx2)-voltage-sensor domain II (VSD2)-voltage-gated sodium (NavAb) channel
  • as an additive in fluorescence quenching experiments of pentaene-SM 18
  • to form isotropic bicelles with p75 for nuclear magnetic resonance (NMR) studies

Biochem/physiol Actions

Facades can be used for the crystallization of membrane proteins since they are considered as optimized membrane mimetics.
