Specificity

Clone 1B15 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects MLH1. It targets an epitope within 22 amino acids from the C-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 22 amino acids from the C-terminal half of human DNA mismatch repair protein MLH1.

Application

Quality Control Testing

Evaluated by Western Blotting in MOLT4 cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected MLH1 in MOLT4 cell lysate.

Tested applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected MLH1 in U2OS cell lysate.

Affinity Binding Assay: A representative lot of this antibody bound MLH1 peptide with a KD of 1.0 x 10-12 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected MLH1 in human colon tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected MLH1 in U2OS cells.

Target description

DNA mismatch repair protein Mlh1 (UniProt: P40692; also known as MutL protein homolog 1) is encoded by the MLH1 (also known as COCA2) gene (Gene ID: 4292) in human. DNA mismatch repair (MMR) is an evolutionarily conserved process that corrects DNA polymerase errors during replication to maintain genomic integrity. MLH1 is a component of the DNA MMR complex composed at least of MSH2, MSH3, MSH6, PMS1 and MLH1. It can form heterodimers with PMS2, PMS1, or MLH3. These heterodimers bind to mismatches and insertions in DNA, as well as certain types of DNA adducts that are induced by drugs, such as cisplatin or monofunctional alkylating agents. The attachment of the MLH1/PMS2 complex is reported to cause degradation of the mutated DNA sequence fragment and the restart of synthesis. MLH1 expression is observed in colon, lymphocytes, breast, lung, spleen, testis, prostate, thyroid, gall bladder, and heart tissue. It plays a key role in correcting mismatches in DNA occurring during DNA replication and have been implicated in the engagement of apoptosis induced by a number of cytotoxic anticancer agents. Mutations in MLH1 gene have been linked to familial predisposition to early-onset colorectal carcinoma (CRC) and extra-colonic tumors of the gastrointestinal, urological, and female reproductive tracts. This ZooMAbZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
