Application

Quality Control Testing

Evaluated by Western Blotting on Pepsin digest of Chicken sternal collagen Type II.

Western Blotting Analysis (WB): A 1:1,000 dilution of this antibody detected Pepsin digest of Chicken sternal collagen Type II.

Tested applications

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Collagen Type II in Human chondrocytes.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Collagen Type II in Human cartilage tissue sections.

Flow Cytometry Analysis: 1 μg from a representative lot detected Collagen Type II in one million Human chondrocytes.

Enzyme Immunoassay Analysis (ELISA): Serial dilutions from a representative lot detected purified Human Collagen Type II.

Target description

Collagen alpha-1(II) chain (UniProt: P02458; also known as Alpha-1 type II collagen) is encoded by the COL2A1 gene (Gene ID: 1280) in human. Collagen is the most abundant protein in the body that is predominantly synthesized by fibroblasts. However, epithelial cells may also synthesize small amounts of collagen. The collagen super family includes over 20 different types of collagens with at least 38 distinct polypeptide chains. Collagens contain a Gly-X-Y repeating structure. The glycine residue is necessary in every third position to ensure close packaging. Although any amino acid can be present at position X or Y, proline is more frequently found in the X position and hydroxyproline in the Y position. Both proline and hydroxyproline stabilize the helical conformation of polypeptide chains. This triplet of amino acids allows collagen chains to twist into a helical structure ( -helix) forming a super helix. Each collagen molecule contains 3 chains twisted around each other to form a triple helix. These 3 chains may either be identical or different depending on the type of collagen. Type II collagen is a major constituent of cartilaginous tissues, representing 90 95% of its total collagen content and forming the fibrils that give cartilage its tensile strength. It is essential for the normal embryonic development of the skeleton, for linear growth and for the ability of cartilage to resist compressive forces. It is synthesized with a signal peptide (aa 1-26) and a propeptide (aa 26-181), which are subsequently cleaved off to produce mature collagen. Cleavage at 181-182 residues is brought about by N-endopeptidases. Excessive cleavage of type II collagen by collagenases is observed in cases of osteoarthritis (OA). This ZooMAbZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
