General description

Transportin 2 (TNPO2 or TRN2) is a member of the karyopherin family of transport receptors. This protein exists in two isoforms TRN2a and TRN2b. The TNPO2 gene is located on human chromosome 19p13.2.

Specificity

Anti-TNPO2 (C-terminal) recognizes human TNPO2 isoform b.

Biochem/physiol Actions

Transportin 2 (TNPO2) is involved in a novel nuclear protein import and export pathway. This protein specifically binds to M9-containing proteins such as heterogeneous nuclear ribonucleoprotein A1 (hnRNPA1) as well as the nucleocytoplasmic shuttling sequence (HNS) region of HuR, but not to the classical nuclear localization signal (NLS)-containing proteins. TNPO2 protein interacts with a similar set of RNA-binding proteins in an RAs-related nuclear protein (Ran)- guanosine triphosphate (GTP) sensitive manner.

Physical form

Solution in 0.01 M phos­phate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

Store at –20 °C. For continuous use, the product may be stored at 2–8 °C for up to one month. For extended storage, freeze in working aliquots at –20 °C. Repeated freezing and thawing, or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
