General description
Protein Kinase C (PKC) is a family of serine/threonine (Ser/Thr)-specific protein kinases. Protein kinase Cε (PKCε) is a member of the novel subgroup. The PKCε isoenzyme appears to be widely expressed in rat brain, lung, skin, liver, spleen, thymus, embryonic rat fibroblasts and in low levels, in certain mouse fibroblasts.
Specificity
Anti-Protein Kinase C ε (PKC ε) reacts in immuno-blotting with PKC ε (90 kDa polypeptide) from rat brain extract
Immunogen
synthetic peptide, corresponding to the C-terminal variable (V5) region (amino acids 726-737) of PKC ε.
Application
Anti-Protein Kinase C epsilon antibody may be used:
- for detection of PKCepsilon in whole cell or membrane extracts of HUVEC cells when stimulated with PMA
- in indirectdot blot
- for protein microarray
Anti-Protein Kinase Cε antibody produced in rabbit may be used in:
- western blot
- immunoprecipitation
- enzyme linked immunosorbent assay (ELISA)
- immunohistochemistry
- chemiluminescence detection systems
Biochem/physiol Actions
Protein Kinase C perform key functions in numerous signalling pathways, in biological systems, through their various isoforms. The PKC isoforms are activated by phosphatidylserine, calcium or phorbol esters. Proteolysis of PKC in vivo is thought to be mediated by calpains I and II. Multiple functions such as, cellular and vascular regulations, angiogenesis, cell growth, apoptosis, changes in basement membrane thickness, extracellular matrix organisation, MAPK signalling, are attributed to PKC isoforms. PKCepsilon and PKCδ have sometimes opposite effects. PKCepsilon is most important in stimulation of cell growth and differentiation and modulation of cytoskeleton These varied functions implicate PKC isoforms in cardiac hypertrophies and diabetic nephropathy and cardiovascular complications.
Protein kinase Cε (PKC epsilon) is expressed in low levels in several normal murine tissues with the exception of brain but is expressed at high levels in several hemopoietic cell lines and tumors, suggesting its possible role in tumorigenesis. Overexpression and stimulation of PKC ε causes an increased growth rate and higher cell densities in monolayers cultures.
Physical form
Rabbit Anti-Protein Kinase C ε is supplied as a liquid containing 0.1% sodium azide as preservative.
Storage and Stability
For continuous use, store at 2–8 °C for up to one month. For extended storage freeze in working aliquots. Repeated freezing and thawing is not recommended. Storage in "frost-free" freezers is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use.
