General description

MMP12 (matrix metalloproteinase 12) is a member of the MMP protein family and is localized to human chromosome 11q22.2-22.3. It is made of nine introns and ten exons, and spans 13kb. It is expressed in a very narrow range of human tissues, and assay of multiple normal human tissues showed its mRNA expression only in placenta. It is found to be expressed in alveolar macrophages of cigarette smokers. It is also called macrophage metalloelastase, and is produced in an inactive form of 54kDa. It is cleaved into an active form of 45kDa, which if further cleaved into 22kDa form. Predominantly, it acts on elastin, and can also degrade fibronectin, laminin, vitronectin etc.

Application

Anti-MMP12 antibody produced in chicken is suitable for western blotting analysis at a dilution of 1:500, for tissue or cell staining at a dilution of 1:200.

Biochem/physiol Actions

Matrix metalloproteinase-12 (MMP-12) may act as a specific marker of Stanford-A acute aortic dissection (AAD). The gene is expressed in the primary tumor and acts as a molecular marker that may be useful for predicting extracapsular spread (ECS) in head and neck squamous cell carcinomas (HNSCC) patients with metastatic nodal disease. It may be associated with asthma through inappropriate attraction of leukocytes to the inflamed tissue. It may also serve as a potential diagnostic tool and a therapeutic target in vascular injury and repair.

Physical form

Solution in phosphate buffered saline containing 0.02% sodium azide.
