General description

DNA damage-inducible transcript 3 protein (UniProt P35638; also known as CCAAT/enhancer-binding protein homologous protein, C/EBP-homologous protein 10, C/EBP zeta, CHOP, CHOP-10, DDIT-3, Growth arrest and DNA damage-inducible protein GADD153) is encoded by the DDIT3 (Also known as CEBPZ, CHOP, CHOP10, GADD153) gene in human (Gene ID 1649). CHOP is a proapoptotic unfold protein response (UPR) gene and a well established marker for ER stress and UPR activation. The accumulation of unfolded or misfolded proteins in the ER induces the activation of three main UPR pathways, the inositol requiring enzyme-1 (IRE1) pathway, the activating transcription factor 6 (ATF6) pathway, and the protein kinase-like ER kinase (PERK) pathway. The PERK pathway involves PERK auto-phosphorylation/activation, resulting in PERK phosphorylation of eIF2α, which in turn leads to increased ATF4 transcription and ATF4-mediated CHOP upregulation.

Specificity

Clone 5E5.3 targeted region is present in both human GADD153/DDIT3/CHOP spliced isoforms reported by UniProt (P35638).

GST-tagged recombinant human GADD153/DDIT3/CHOP internal fragment.

Application

Immunocytochemistry Analysis: 5.0 µg/mL from a representative lot detected GADD153/DDIT3 in HeLa cells.
Immunohistochemistry Analysis: A 1:50 dilution from a representative lot detected GADD153/DDIT3 in human brain, kidney, and skeletal muscle tissue sections.

This Anti-GADD153 Antibody, clone 5E5.3 is validated for use in Western Blotting, Immunocytochemistry, Immunohistochemistry (Paraffin) for the detection of CHOP.

Quality

Evaluated by Western Blotting Analysis in HUVEC lysate.

Western Blotting Analysis: 1.0 µg/mL of this antibody detected GADD153/DDIT3 in 10 µg of HUVEC lysate.

Target description

~20 kDa observed. 19.18 kDa (isoform 1) and 21.70 kDa (isoform 2) calculated. Uncharacterized band(s) may appear in some lysates.
