General description

Platelet-derived growth factor (PDGF) is a 30 kDa cationic polypeptide and consists of two disulphide linked chains A and B. PDGF is expressed as hetero and homodimers : PDGF-AB FDGF-AA, PDGF-BB, PDGF-CC and PDGF-DD.

Application

Platelet-Derived Growth Factor-BB from rat has been used as a growth supplement in mouse embryonic fibroblast culture, motoneuron-muscle co-culture and adult spinal cord neurons culture.

Biochem/physiol Actions

Platelet-derived growth factor (PDGF) elicits multifunctional actions with a variety of cells. It is mitogenic to mesoderm-derived cells, such as dermal and tendon fibroblasts, vascular smooth muscle cells, glial cells, and chondrocytes. PDGF is a potent chemoattractant and activator of neutrophils, monocytes, and fibroblasts. PDGF activates smooth muscle relocalization and multiplication. PDGF mediated increased smooth muscle muscle proliferation might be associated with gene regulation. It increases the synthesis of phospholipids, cholesterol esters, glycogen and prostaglandins, and modulates LDL (low density lipoprotein) receptor binding.

Physical form

Lyophilized from a 0.2 μm filtered solution in 30% acetonitrile and 0.1% trifluoroacetic acid containing 2.5 mg bovine serum albumin.

Analysis Note

The biological activity is measured by its ability to stimulate 3H-thymidine incorporation in quiescent NR6R-3T3 fibroblasts.
