General description

Cullin-1 (Cul1) is encoded by the gene mapped to human chromosome 7q36.1. It is a 85 kDa protein that belongs to the family of cullin protein family containing distinct group of mediators (at least eight cullin members Cul1-8).

Immunogen

synthetic peptide located at the C-terminus of human cullin-1 (amino acids 759-776). This sequence is identical in mouse cullin-1 and highly conserved (83% identity) in Drosophila cullin-1.

Application

Anti-Cullin-1 antibody produced in rabbit has been used in western blotting.

Biochem/physiol Actions

Cullin 1(CUL1) is a scaffold protein of the ubiquitin E3 ligase Skp1/Cullin1/Rbx1/F-box protein complex, which ubiquitinates a broad range of proteins that are involved in cell-cycle progression, signal transduction and transcription. Cul1 plays an important role in protein degradation and protein ubiquitination. It regulates breast cancer cell proliferation, migration and invasion. Overexpression of Cul1 leads to breast cancer progression. CUL1 promotes human trophoblast cell invasion and dysregulation of its expression may be associated with pre-eclampsia (PE). It may have some role in rheumatoid arthritis (RA) and its response to methotrexate. Cullin1 may act as an important marker for human gastric cancer lymph node metastasis and prognosis.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 1% bovine serum albumin and 15 mM sodium azide.
