General description

The TrueGel3D hydrogel pre-configured kit is used to prepare a chemically defined hydrogel with stiffness of ~400 Pa (shear modulus) that can be customized to match that of native cell environment. The TrueGel3D hydrogel is formed by crosslinking of RGD (arginylglycylaspartic acid) degradable polymer with CD cell-degradable crosslinker. The cells are encapsulated during crosslinking, where they can adhere to the polymer through the RGD peptide and grow within the hydrogel. The CD cell-degradable crosslinker is composed of matrix metalloprotease (MMP)-cleavable peptide (Pro-Leu-Gly-Leu-Trp-Ala), which allows cells to spread and migrate by secreting matrix metalloproteases (MMP1, MMP3, MMP7 and MMP9).

Gel can be further dissolved by TrueGel3D Enzymatic Cell Recovery Solution (TRUEENZ-500UL) for post-culture analysis including RT-PCR, western blots and other molecular biological studies.

Application

Suitable for 3D-culture of various cell types, spheroids and tissue samples. It has been used for cell migration and invasion studies, co-culture experiments (tumor cells with fibroblasts) and cell growth and differentiation studies.

Components

TRUE1-1KT contains lyophilized RGD degradable polymer and CD cell-degradable crosslinker with reconstitution buffer and water.

Caution

Avoid prolong exposure of CD cell-degradable crosslinker to air.
