General description

La mitomicina C es un quimioterápico sistémico y un antibiótico obtenido de Streptomyces caespitosus.[1][2] Es el primer alquilante biorreductor que se identificó y es un derivado del ácido 3-amino-5-hidroxibenzoico (AHBA), la D-glucosamina, la L-metionina y el carbamoilfosfato.[3]

Application

La mitomicina C de Streptomyces caespitosus se ha utilizado para el tratamiento de capas de células alimenticias, como los PMEF (fibroblastos embrionarios primarios de ratón) y los fibroblastos embrionarios de ratón CD1 (MEF) para el cultivo de hESC (células madre embrionarias humanas).[4][5][6] También se ha utilizado para el tratamiento de la línea celular BLC (cáncer «basal-like»).[7]

Biochem/physiol Actions

La mitomicina C provoca el entrecruzamiento del ADN bicatenario, lo que produce mutagénesis, inhibición de la síntesis de ADN, inicio de eventos de reparación del ADN y activación de la apoptosis. La capacidad para inhibir la síntesis de ADN es atribuible al grupo aminoquinona presente en la mitomicina C.[3][8] Este compuesto no afecta a la síntesis de ARN ni de proteínas. En cultivo de tejidos, este compuesto reduce la viabilidad celular y suprime la mitosis, y causa desorganización del núcleo y la producción de células gigantes.[2] La mitomicina C tiene potentes actividades anti-gramnegativas y anti-grampositivas.[8] Contiene tres restos, los grupos quinina, uretano y aciridina. Se utiliza para generar células nutritivas mitóticamente inactivas en sistemas de cultivo celular, como los fibroblastos mitóticamente inactivos utilizados en los sistemas de células madre embrionarias.
Modo de acción: Este producto es un agente alquilante dirigido específicamente a la secuencia del nucleósido de guanina 5′-CpG-3′. Inhibe la síntesis de ADN mediante reacción covalente con el ADN al establecer enlaces transversales entre las cadenas complementarias del ADN. Esta interacción evita la separación de las cadenas de ADN complementarias, inhibiendo así la replicación del ADN.

Espectro antimicrobiano: La mitomicina C tiene una fuerte actividad antitumoral, en especial contra las células tumorales de la ascitis de Ehrlich, y una fuerte acción bactericida contra las bacterias grampositivas y gramnegativas.

Caution

Este vial contiene 2 mg de mitomicina C y 48 mg de NaCl. Las disoluciones de reserva deben esterilizarse mediante filtración y conservarse a 2 - 8°C en la oscuridad. Las disoluciones a pH 6-9 pueden conservarse entre 0 y 5°C durante un máximo de una semana, pero si se forma un precipitado, debe prepararse una nueva disolución; se ha demostrado que la disolución precipitada es tóxica para las células.

Preparation Note

La mitomicina C es soluble en agua a 0,5 mg/ml, con un pH de 6,0-7,5. Experimenta degradación rápida en disoluciones ácidas con un pH<6 y es más probable que conserve su actividad en disoluciones con un pH entre 6 y 9.
