General description

Tn antigen is a truncated O-glycan, that is commonly noticed in colorectal cancer (CRC). It is usually expressed at high levels in cancer cells. Tn antigen is small in size and simple in structure. It has a non-physiological glycan structure in humans, hence it may be identified as foreign by the immune system.

Biochem/physiol Actions

Tn antigen stimulates colorectal cancer metastasis through the activation of epithelial-mesenchymal transition, mediated by H-Ras. It acts as a precursor to several significant complex O-glycans. Subpopulations of blood cells in all lineages carrying the Tn antigen results in a hematological disorder called Tn syndrome.

Colorectal cancer is the third most common cancer type worldwide. It is characterized by a high expression of aberrantly glycosylated ligands, such as the Tn antigen (GalNAcα1-Ser/Thr), which is a major ligand for the C-type lectin macrophage galactose-type lectin (MGL).

Mucin-type O-linked glycosylation, a posttranslational modification affecting the stability and biophysical characteristics of proteins, requires C1GalT1 (T synthase) and its obligate, X-linked chaperone Cosmc. Hypomorphic C1GalT1 mutations cause renal failure via not yet established mechanisms. We hypothesize that impaired Cosmc-dependent

Immature truncated O-glycans such as Tn antigen are frequently detected in human colorectal cancer (CRC); however, the precise pathological consequences of Tn antigen expression on CRC are unknown. T-synthase is the key enzyme required for biosynthesis of mature O-glycans. Here

Pancreatic adenocarcinoma (PDAC) is a leading cause of cancer-related death. Altered glycosylation contributes to tumor progression and chemoresistance in many cancers. C1GALT1 is the key enzyme controlling the elongation of GalNAc-type O-glycosylation. Here we showed that C1GALT1 was overexpressed in
