General description

Dynamin-1 & Dynamin-2 belong to the dynamin superfamily of large GTPases. Dynamins are multidomain proteins consisting of a GTP-binding domain, a middle domain, and a GTP-effector domain. Dynamins also contain a pleckstrin homology (PH) domain which may facilitate membrane binding. Dynamin-1 is highly abundant in the brain particularly at synaptic contacts, whereas Dynamin-2 is ubiquitously expressed in non-neuronal cells. Dynamins are involved in receptor-mediated endocytosis of clathrin-coated vesicles. Dynamin-1 interacts with an array of cellular targets via its SH3 binding domain and is implicated in late stages of vesicle formation by forming oligomers around the “neck” of budding vesicles and inducing membrane fission; it may also play a role in cytoskeleton rearrangement and cytoskinesis. Dynamin-2 is involved in the formation of clathrin-coated vesicles but may also be involved in the recycling of receptors between endosomes and the trans-Golgi network.

Recombinant full length human Dynamin-1 expressed in transfected Sf9 cells.
