General description

Signal transducer and activator of transcription 2 (UniProt: Q9WVL2; also known as STAT2) is encoded by the Stat2 gene (Gene ID: 20847) in murine species. The JAK-STAT signaling pathway play an important role in cell proliferation, cell differentiation, cell migration, and cell death. It is the principal signaling mechanism for a variety of cytokines and growth factors. In mammals, seven different STATs have been recognized. They contain a conserved tyrosine residue near the C-terminus, which is phosphorylated by JAKs, which allows the dimerization of STATs by interacting with a conserved SH2 domain. In their latent form STATs are located in the cytoplasm and upon phosphorylation they are transported into the nucleus by an importin -5 dependent mechanism. In the nucleus, dimerized STATs bind to specific regulatory sequences to activate or repress transcription of target genes. The main function of STAT2 is to mediate IFN- /β signaling. Following type I interferon binding to cell surface receptors, JAK kinases are activated, leading to tyrosine phosphorylation of STAT1 and STAT2. The phosphorylated STATs dimerize, associate with IRF9/ISGF3G to form a complex termed ISGF3 transcription factor, that enters the nucleus. ISGF3 binds to the IFN stimulated response element (ISRE) to activate the transcription of interferon stimulated genes. STAT2 is also reported to serve as a regulator of mitochondrial fission by modulating the phosphorylation of DNM1L at serine 616 and serine 637, which activate and inactivate the GTPase activity of DNM1L respectively. Specificity

This rabbit polyclonal antibody detects STAT2 phosphorylated on tyrosine 689.

Immunogen

KLH-conjugated linear peptide corresponding to 9 amino acids surrounding phosphotyrosine 689 from the C-terminal half of mouse STAT2.

Application

Tested Applications
  • Immunoprecipitation Analysis: 10 µg from a representative lot immunoprecipitated phospho-STAT2 (Tyr689) in lysate from HeLa cells stimulated with Interferon and Interferon
  • Peptide Inhibition Assay: Target band detection in lysate from HeLa cells stimulated with Interferon and Interferon was prevented by pre-blocking of a representative lot with the immunogen phosphopeptide, but not the corresponding non-phosphopeptide.
  • Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
Anti-phospho-STAT2 (Tyr689), Cat. No. 07-224, is a rabbit polyclonal antibody that detects STAT2 phosphorylated on tyrosine 689 and is tested for use in Immunoprecipitation, Peptide Inhibition, and Western Blotting.

Quality

Evaluated by Western Blotting in HeLa cells stimulated with Interferon and .Western Blotting Analysis (WB): A 1:200 dilution of this antibody detected STAT2 phosphorylated on tyrosine 689 in HeLa cells stimulated with Interferon (0.3 ng/mL for 16 h) and Interferon ( 2000 U/mL for 20 min).

~110 kDa observed; 105.42 kDa calculated. Uncharacterized bands may be observed in some lysate(s).
