General description

Histone H3 is one of the five main histone proteins involved in the structure of chromatin in eukaryotic cells. Featuring a main globular domain and a long N-terminal tail, H3 is involved with the structure of the nucleosomes of the ′beads on a string′ structure. The N-terminal tail of histone H3 protrudes from the globular nucleosome core and can undergo several different types of epigenetic modifications that influence cellular processes. These modifications include the covalent attachment of methyl or acetyl groups to lysine and arginine amino acids and the phosphorylation of serine or threonine. Histone H3 variants (H3.1, H3.2 and H3.3) have been implicated in the epigenetic memory of cellular state. Genome-wide patterns of H3 are dependent on amino acid sequence and change with cellular differentiation at developmentally regulated loci.

Immunogen

KLH-conjugated synthetic peptide (ARTKQTARK(Ac)STG-C) corresponding to amino acids 1-12 of histone H3 with acetylated Lys9.

Application

Anti-acetyl-Histone H3 (Lys9) Antibody, Alexa Fluor 555 Conjugate, Cat. No. 06-942-AF555, is a highly specific rabbit polyclonal antibody that targets histone H3 Lys9 acetylation (H3K9ac) and has been tested in Flow Cytometry and Immunocytochemistry.
Flow Cytometry Analysis: 2 µL from a representative lot detected cell population with histone H3 Lys9 acetylation (H3K9ac) among one million HEK293 cells.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected nuclear histone H3 Lys9 acetylation (H3K9ac) in A431, HeLa, and NIH/3T3 cells.

The unconjugated antibody (Cat. No. 06-492) is shown to be suitable also for Western blotting, dot blot, flow cytometry, chromatin immunoprecipitation (ChIP) and ChIP-seq applications.

Quality

Evaluated by Immunocytochemistry in HUVECs.

Immunocytochemistry Analysis: A 1:100 dilution of this antibody detected nuclear histone H3 Lys9 acetylation (H3K9ac) in HUVECs.
